La pregabalina es un análogo del GABA, asociado con la gabapentina, y se ha estudiado ampliamente en el tratamiento del trastorno de ansiedad generalizada, la fibromialgia, la neuralgia y la epilepsia parcialmente compleja. Parece actuar de forma más selectiva sobre las subunidades α2-δ de los canales de calcio cerebrales que la gabapentina. La pregabalina fue aprobada por la FDA a finales de 2004 para el tratamiento del dolor neuropático y la epilepsia. En 2008, se convirtió en el primer fármaco aprobado para el tratamiento de la fibromialgia. Si bien la pregabalina fue aprobada en la Unión Europea en 2006 para el tratamiento del trastorno de ansiedad generalizada, al momento de redactar este documento, la FDA no ha aprobado su uso en trastornos de ansiedad generalizada en Estados Unidos. Debido a la eficacia consistente de la pregabalina en ensayos clínicos y al perfil de seguridad conocido en la experiencia clínica, no está del todo claro qué impide su aprobación para el tratamiento del trastorno de ansiedad generalizada. Se han reportado casos de riesgo-beneficio del fármaco, dada la hepatotoxicidad que se presenta en microbios, pero no se ha demostrado que esto represente un problema tras su liberación en humanos. La pregabalina está clasificada como anticonvulsivante y analgésico. El fármaco se encuentra actualmente bajo revisión por la FDA. La pregabalina presenta potencial de abuso y uso indebido. Está clasificada como un compuesto de Clase V en Estados Unidos.

Pregabalin is an analogue of GABA, associated with gabapentin, and has been extensively studied in the treatment of generalized anxiety disorder, fibromyalgia, neuralgia, and partially complex epilepsy. It appears to act more selectively on the a2-δ subunits of calcium channels in the brain than gabapentin. Pregabalin was approved by the FDA in late 2004 for the treatment of neuropathic pain and epilepsy. In 2008, it became the first drug approved for the treatment of fibromyalgia. Although pregabalin was approved in the European Union in 2006 for the treatment of generalized anxiety disorder, at the time of writing, the FDA has not approved pregabalin for use in generalized anxiety disorders in the United States. Because of the consistent efficacy of pregabalin in clinical trials and the known safety profile in clinical experience, it is not entirely clear what is preventing the approval of pregabalin for the treatment of generalized anxiety disorder. There have been reports that the risk-benefit ratio of the drug is worrying given the hepatotoxicity that occurs in micc, but this has not been proven to be a problem after human release. Pregabalin is classified as an anticonvulsant and analgesic. The drug is currently under review by the FDA. Pregabalin has the potential for abuse and misuse. Pregabalin is classified as a Class V compound in the United States.
